In the summer the Kegen border crossing is open and you can research how to cross that border as there are a lot of posts here about it.

In the winter the fastest and easiest thing to do is probably backtrack to Almaty and then go to Bishkek and Karakol from there.

They usually don't care about the visa of the first country, as if you can't get a visa for that country there's no way for you to actually use the transit visa.

In addition to the above, this only comes close to being realistic if you have hired a vehicle beforehand and are doing everything in that vehicle. Even so, you don't have much buffer time in case anything goes wrong.

The road doesn't have scheduled closures and isn't seasonal, but I assume like all roads it may be closed or impassable after heavy snow.

That person will be basically be responsible for you the entire time you're in Turkmenistan. Is that a responsibility they've considered and are willing to accept and/or one you're willing to place on them?
