There's (currently) a night train on Wednesdays and Fridays from Bishkek (20:23) to Samara that stops in Shymkent (arrival at 11:37) - this would save you the trouble of going back to Almaty for the night train experience. (If you go for this, please re-check times as different websites show schedules from/to Central Asian cities based on different time zones and in different ways. For example, a departure time may be displayed in Moscow time while the arrival time is in local time - or vice versa... This particularly seems to be the case for trains going to Russia.)
Turkestan is "as Uzbekistan as it gets" in Kazakhstan, but it is a far cry from the real thing in Samarkand and other places on the Silk Road. There are quite a few trains between Turkestan and Shymkent and they take at least three hours. The night train from Bishkek also stops in Turkestan.
